SLU-PP-332 is a small molecule agonist of estrogen-related receptors alpha and gamma (ERRα and ERRγ), identified through structure-based virtual screening at Saint Louis University by Bhatt, Bhatt, and colleagues. Despite their name, estrogen-related receptors (ERRs) are orphan nuclear receptors that do not bind estrogens — they are constitutively active transcription factors that regulate gene programs controlling mitochondrial biogenesis, oxidative phosphorylation, fatty acid beta-oxidation, and muscle fiber type specification.

The mechanism of SLU-PP-332 involves enhancing ERR transcriptional activity, upregulating PGC-1α (peroxisome proliferator-activated receptor gamma coactivator 1-alpha) target gene programs that drive a slow-twitch, oxidative muscle fiber phenotype. In skeletal muscle research models, ERR activation promotes expression of type I fiber genes (myoglobin, slow myosin heavy chain), increases mitochondrial DNA copy number and citrate synthase activity (markers of mitochondrial content), enhances muscle capillary density (angiogenic remodeling), and improves fatigue resistance. This comprehensive metabolic reprogramming produces a cellular and tissue phenotype that closely recapitulates the adaptations produced by endurance exercise training — establishing SLU-PP-332 as a pharmacological exercise mimetic.

Published in Cell Metabolism and other high-impact journals, SLU-PP-332 research has demonstrated increased running endurance and reduced fatigue in animal models without exercise training, making it a tool for dissecting exercise adaptation mechanisms.
