Human Chorionic Gonadotropin (HCG) is a heterodimeric glycoprotein hormone consisting of a 92-amino acid alpha subunit (shared with LH, FSH, and TSH) and a 145-amino acid beta subunit unique to HCG that confers receptor specificity. The molecule is heavily glycosylated, with carbohydrate moieties comprising approximately 30% of its molecular mass, contributing to its extended biological half-life in research systems compared to the structurally related luteinizing hormone (LH).

HCG binds to and activates the luteinizing hormone/chorionic gonadotropin receptor (LHCGR), a G-protein coupled receptor (GPCR) expressed predominantly in gonadal tissue. In Leydig cells (testicular interstitial cells), LHCGR activation stimulates cAMP/PKA signaling that drives cholesterol transport into mitochondria via StAR (steroidogenic acute regulatory) protein, initiating the steroidogenic cascade that converts cholesterol to testosterone through sequential enzymatic steps (CYP11A1, CYP17A1, HSD3B, HSD17B). In ovarian tissue, LHCGR activation drives theca cell androgen production, corpus luteum maintenance, and progesterone synthesis. The longer half-life and greater receptor affinity of HCG compared to LH make it the preferred tool in gonadal function research.

Research applications include Leydig cell steroidogenesis studies, ovarian follicular dynamics, corpus luteum biology, gonadotropin receptor pharmacology, and reproductive endocrinology.
