FSH acts primarily through the FSH receptor (FSHR, Gs-coupled GPCR) on granulosa cells (ovary) and Sertoli cells (testis), driving follicular development, aromatase expression, inhibin production, and spermatogenic support. LH acts through the LHCGR on theca cells and Leydig cells, driving androgen production that serves as the aromatase substrate for estrogen synthesis and independently maintaining steroidogenic output. The two-cell, two-gonadotropin model of ovarian function — in which LH-driven theca cell androgens are converted to estrogens by FSH-stimulated granulosa cell aromatase — can only be fully studied with preparations providing both activities.
