Pinealon (EDR) is a synthetic tripeptide bioregulator targeting pineal gland function and neuroprotection. Part of the Khavinson short peptide bioregulator series studied for epigenetic gene regulation in aging research.

Pinealon (sequence: Glu-Asp-Arg, abbreviated EDR) is a synthetic tripeptide bioregulator developed by Professor Vladimir Khavinson at the St. Petersburg Institute of Bioregulation and Gerontology as part of a comprehensive research program investigating tissue-specific short peptide signaling molecules. The bioregulator concept proposes that short peptides (2-4 amino acids) serve as endogenous epigenetic modulators that interact with specific DNA sequences in gene promoter regions to regulate tissue-specific gene expression, representing a layer of biological regulation between classical hormone signaling and direct transcription factor activity.

Published research on Pinealon has focused on its interaction with pinealocyte and neuronal cell biology. Cell culture studies report that Pinealon is capable of penetrating cell membranes and nuclear envelopes without requiring receptor-mediated endocytosis — a property attributed to its small size and specific charge distribution. Once nuclear, the peptide is proposed to interact with histone proteins and specific DNA sequences to modulate chromatin accessibility and gene transcription. In pinealocyte models, Pinealon has been studied for its effects on melatonin synthesis pathway gene expression, while in neuronal models, research has focused on neuroprotection against oxidative stress, hypoxia-induced damage, and excitotoxicity.

Additional research applications include age-related cognitive decline models, circadian rhythm regulation studies, and comparative bioregulator research (Pinealon vs. Epithalon vs. other Khavinson peptides). The tripeptide has been the subject of multiple publications from the St. Petersburg Institute, with independent investigations beginning to emerge from other research groups interested in the short peptide bioregulation paradigm.
